LHX2 is a transcription factor of the LIM/homeobox family of proteins. It has an essential role in the specification of the posterior lobe and controlling of anterior pituitary development. Loss of LHX2 leads to anterior lobe agenesis with a subsequent phenotype of anterior pituitary hypoplasia and hyperphagia. We identified a missense mutation of the LHX2 gene in a Turkish family with two affected individuals with an unusual phenotype of bilateral posterior lobe agenesis, pituitary hypoplasia and hyperphagia. The genotype-phenotype relationship of LHX2 variants is still unknown. The purpose of this study is to delineate the effect of this LHX2 mutation on the human phenotype. A novel mutation c.1053G > A (p.S351T) of LHX2 was found in a Turkish family with three affected individuals with an unusual phenotype of bilateral posterior lobe agenesis, pituitary hypoplasia and hyperphagia. Alignment of the wild-type and mutant amino acids sequences was performed by UniProtKB. The functional consequences of the identified mutation were evaluated by in silico methods. This variant causes the loss of the highly conserved amino acids serine in position 351. This mutation is most likely the cause of the phenotype in this family. In silico analysis indicates that the mutation is damaging and is probably the cause of the phenotype in this family. The novel mutation of the LHX2 gene reported in this study is probably the cause of the phenotype in the present family.
